  • CD98hc as a marker of radiotherapy-resistant cancer stem cells in head and neck squamous cell carcinoma

    <jats:sec><jats:title>Introduction</jats:title><jats:p>The prognosis of head and neck squamous cell carcinoma (HNSCC) is generally good in cases of high radiation sensitivity but poor in cases exhibiting radiation resistance; this resistance has been attributed to the presence of cancer stem cells. In recent years, CD98hc overexpression has been associated with poor prognosis in various types of cancers. CD98 is a heterodimer of heavy and light chains and is strongly involved in cell proliferation, survival, migration, and adhesion. We investigated whether CD98hc can be used as a cancer stem cell marker for HNSCC.</jats:p></jats:sec><jats:sec><jats:title>Material and methods</jats:title><jats:p>We exposed five HNSCC cell lines to a total radiation dose of 60 Gy administered in 2 Gy fractions on consecutive days to investigate changes in CD98hc expression. Furthermore, we separated CD98-positive and CD98-negative cell populations to comparatively investigate the properties of each.</jats:p></jats:sec><jats:sec><jats:title>Results</jats:title><jats:p>Radiation resistance was observed in all five cell lines, and resistant cells exhibited CD98hc overexpression, with enhanced spheroid formation, migratory, and invasive abilities. Radiation-resistant cell lines were separated into CD98-positive and CD98-negative populations. CD98hc-positive radiation-resistant cell lines exhibited enhanced spheroid formation, invasion, and plating efficiency as well as strong tumorigenicity in nude mice.</jats:p></jats:sec><jats:sec><jats:title>Conclusions</jats:title><jats:p>CD98hc-positive cells exhibited cancer stem cell-like properties in all cell lines. Thus, CD98hc is a potential marker of radiation sensitivity as well as a potential therapeutic target for improved survival rates of patients with HNSCC.</jats:p></jats:sec>

  • Amyloid β-related angiitis presented only a leptomeningeal legion and enlarged perivascular spaces without detection of hemorrhagic lesions at first visit: a case report

    <p>A 70-year-old woman visited our hospital with a chief complaint of headache. T2-weighted and fluid-attenuated inversion-recovery magnetic resonance imaging (MRI) showed a leptomeningeal lesion in the right temporal lobe and many enlarged perivascular spaces in the centrum semiovale. Follow-up MRI performed 6 months later revealed cortical microbleeds and cortical superficial siderosis. Three years later, the patient presented with right lower quadrant homonymous hemianopsia and disturbance of consciousness. MRI showed diffuse leptomeningeal and significantly increased hemorrhagic lesions. A brain biopsy confirmed diagnosis of amyloid β-related angiitis. Disturbance of consciousness persisted despite steroid treatment. We concluded that vasculitis-induced inflammation preceded bleeding secondary to vascular fragility in our patient. Inflammatory cerebral amyloid angiopathy should be included in the differential diagnosis in patients with early-onset leptomeningeal lesions and enlarged perivascular spaces without hemorrhagic lesions.</p>

  • Endovascular treatment of acute intracranial large-vessel occlusion caused by a thrombus in the cervical internal carotid artery

    <p>  We describe a case in which a cardiogenic thrombus at the cervical internal carotid artery migrated to the middle cerebral artery. The patient was a 69‒year‒old female who had mild neurological symptoms. At her first visit, magnetic resonance diffusion‒weighted imaging indicated no acute ischemic lesion. Magnetic resonance angiography revealed occlusion of the right internal carotid artery and a patent middle cerebral artery supplied via the posterior communicating artery. The next day, the patient exhibited left hemiparesis and occlusion of both the internal carotid artery and middle cerebral artery. A mechanical thrombectomy was performed. Complete recanalization was achieved, and the patient's neurological deficits were resolved. The mechanical thrombectomy was effective for this emergency state of thrombus migration from the cervical internal carotid artery to the terminus of the internal carotid artery. The present case demonstrates that it is possible for a thrombus of the cervical internal carotid artery to migrate to a distal intracranial artery and result in severe symptoms. In the event of such an emergency, a system for rapid treatment by mechanical thrombectomy is needed.</p>
