  • Morphologic observation for anomalous patterns of the flexor carpi radialis muscle and atypical insertions: a proposal for a new classification

    In the present study, four types of variations of the flexor carpi radialis with accessory muscular bundles were classified depending on the origin of the accessory muscular bundle and relationships with the bicipital aponeurosis, biceps brachii, pronator teres, and coracobrachialis. Six types of insertions of the tendon of the flexor carpi radialis were also divided according to their inserted positions on the carpal and metacarpal bones. An accessory muscular bundle of the flexor carpi radialis was found in 4 (1 female and 3 males) of 68 cadavers (5.88%), with five examples in 136 arms (3.68%). It was bilateral in one cadaver and unilateral (two on the right and one on the left) in three. The insertion of the flexor carpi radialis tendon was not only on the proximal surface of the base of the second metacarpal bone but also on the third metacarpal bone, the tubercle of the trapezium, and the scaphoid. These findings and classifications are important in anatomical education, and have important significances in clinical diagnosis and therapies.

  • Different localization of ATP sensitive K+ channel subunits in rat testis.

    ATP sensitive K+ (KATP) channels are important linkage for cell membrane excitability with the cellular bioenergetic state. These channels are in vivo composed of pore-forming subunits and regulatory subunits. The present study was focused on the cellular expression and localization of these subunits in rat testis. RT-PCR analysis showed that rat testis contains five KATP channel subunits, Kir6.1, Kir6.2, SUR1, SUR2A and SUR2B. Immunoblot assay showed that proteins of Kir6.1, Kir6.2, SUR2A and SUR2B were expressed in rat testis. Immunohistochemistry revealed that the spermatogenic cells, Sertoli cells and Leydig cells show positive staining for these KATP channel subunits with different localizations, implying these subunits play roles during spermatogenesis. A double immunofluorescence labeling study demonstrated that Kir6.1 does not colocalize with SUR2A or SUR2B, but Kir6.2 colocalizes with SUR2B in the head cap or acrosome of spermatids, suggesting that the KATP channel in spermatids is formed by Kir6.2 and SUR2B.

    ATP sensitive K+ (KATP) channel subunits were investigated in rat submandibular gland (SMG). RT-PCR detected the presence of mRNA transcripts of the Kir6.1, Kir6.2, SUR2A and SUR2B in the SMG, while SUR1 mRNA was barely detected. Western blot analysis provided the evidence that these four KATP channel subunits expressed in rat SMG. Immunostaining detected that these four KATP channel subunits are widely distributed in myoepithelial cells, epithelial cells of intercalated ducts, granular convoluted tubules, striated ducts, and excretory ducts with different intensities. Immunofluorescence double staining showed that Kir6.1 or Kir6.2 colocalized with SUR2A in the myoepithelial cells, granular convoluted tubule, striated duct, and excretory duct. Kir6.1 or Kir6.2 also colocalized with SUR2B mainly in the duct system such as the granular convoluted tubules, striated ducts, and excretory ducts. Taken together with these results, the KATP channels in SMG may be consisted of Kir6.1, Kir6.2, SUR2A and SUR2B, with various combinations of colocalization with each other, and may play important roles in rat SMG during the salivary secretion.
